Genotyping Kit - Venous Thrombosis
The Genotyping Kit ? Venous Thrombosis allows the determination of the R/Q 506 mutation in the factor V gene (Factor V Leiden), the 20210 G/A mutation in the prothrombin (Factor II) gene and A/V 223 (677 C/T) mutation in the 5,10-methylene-tetrahydrofolate reductase gene (MTHFR).
The assay consists of three steps:
1. DNA extraction 2. DNA amplification by PCR 3. analysis of PCR products by gel- electrophoresis.
Step (2) is based on a "multiplex" PCR reaction, which combines multiple primer combinations in the same reaction tube. In this kit 6 specific primer combinations are used to amplify mutant as well as wild type (normal variant) alleles of three different genes. Thus, homozygous and heterozygous carriers of the mutations can be identified by one PCR test. The Genotyping Kit contains DNA diluent (sterile 2x3 mL), mastermix for 2x11 PCR reactions(2x360 µL), size marker (1x20 µL), Sybr®Green I (5µL sample), 2 ready to use Spreadex® EL 400 Mini gels, 1 gel frame and the Electrophoresis buffer 40 x concentrated.
